Figure 6.

TNF-stimulated necrosis requires ROS production. (A) Wild-type (WT) and Jnk-/- fibroblasts expressing ΔN-IκBα were treated with 10 ng/mL TNF (12 h). Cell survival was measured by staining with crystal violet. The effect of the addition of butylated hydroxyanisole (BHA; 100 μM) was examined. The data shown are the mean ± SD of triplicate determinations. The data are representative of three independent experiments. (B) Wild-type (WT) and Jnk-/- fibroblasts were incubated (4 h) with different concentrations of hydrogen peroxide. The effect of ΔN-IκBα expression was examined. Cell survival was measured by staining with crystal violet. The data shown are the mean ± SD of triplicate determinations. The data are representative of three independent experiments. (C) Wild-type (WT) (ΔN-IκBα) and Jnk-/- (ΔN-IκBα) fibroblasts were incubated with 50 μM hydrogen peroxide. Cell survival was measured by staining with crystal violet. The data shown are the mean ± SD of triplicate determinations. The data are representative of three independent experiments.
