JNK-dependent ROS production contributes to TNF-stimulated necrosis. (A) Wild-type (WT) and Jnk-/- fibroblasts expressing ΔN-IκBα were incubated (4 h) without or with 10 ng/mL TNF or 10 ng/mL TNF plus 2 μM emetine. The cells were stained with 1 μM CM-H2DCFDA to examine ROS production (upper panels; green) and with DAPI to detect nuclear DNA (lower panels; blue). The data shown are representative of three independent experiments. (B) Jnk-/- (ΔN-IκBα) fibroblasts were stained with CellTracker Red CMTPX, cocultured with wild-type (WT) (ΔN-IκBα) fibroblasts, and incubated (4 h) with 10 ng/mL TNF. The cells were stained with 1 μM CM-H2DCFDA to examine ROS production (green) and with DAPI to detect nuclear DNA (blue). The data shown are representative of three independent experiments.
