(A) qRT-PCR was used to measure the expression level of PVT1 in U2OS and MG-63 cells that had been transfected with control or siRNAs against PVT1, i.e., si-PVT1#1, si-PVT1#2, or si-PVT1#3 (*P < 0.05). (BC) Cell proliferation was assessed with an MTT assay in U2OS and MG-63 cells transfected with control or si-PVT1#1 (*P < 0.05). (DE) Cell apoptosis was assessed by Annexin-V/7-AAD staining in U2OS and MG-63 cells transfected with control or si-PVT1#1 (*P < 0.05). (F) An immunofluorescence assay was performed to measure TUNEL expression in U2OS and MG-63 cells transfected with control or si-PVT1#1. (GH) A clonal colony-forming assay was performed to assess cell proliferation in U2OS and MG-63 cells transfected with control or si-PVT1#1 (*P < 0.05).
