A. IMR-32 cells were treated with the increasing concentrations of SAR405838, Nutlin-3 and RG7388 for 72 hours, respectively. Cell viability was then measured by CCK-8 assay. B. The IC50s of SAR405838, Nutlin-3 and RG7388 in IMR-32 cell line were calculated based on the data collected in the cell viability assay by using Prism 5. C. The IMR-32 cells were treated with an identical 10 μM concentrations of SAR405838, Nutlin-3 and RG7388 for 12 hours. Cells were then lysed and subjected to SDS-PAGE followed by immunoblotting for the antibodies indicated. The α Tubulin antibodies were used as a loading control for whole cell extracts.
