(A, B) Cells were pretreated for 1 h with or without the indicated concentrations of FTY720 and exposed to different dose of radiation. The cells were then cultured for 48 h in the presence of FTY720. Cell viability was measured in CNE-1 (A) and CNE-2 (B) cells using CCK-8 kits. (C) The percentage of apoptotic cells in the cells that were treated with FTY720 (5 μM), radiation (4 Gy), or a combination of the two was measured using flow cytometric analysis. (D) FTY720 plus radiation significantly decreased SPHK1 enzymatic activity in CNE-1 and CNE-2 cells. SPHK1 activity was measured using a Sphingosine Kinase Activity Assay Kit. (E) Western blot analysis of protein expression levels in CNE-1 and CNE-2 cells that were treated with FTY720 (5 μM), radiation (4 Gy), or a combination of the two for 48 h. *P < 0.05.
