A. DMEM medium contained 25 mM glucose (down-pointing triangles; Glc), 3.85 mM glutamine (up-pointing triangles; Gln) or 24.5 mM glucose + 3.85 mM glutamine (circles; Gln+Glc). TMRE fluorescence in the presence of HNE was normalized to the TMRE fluorescence without HNE (insert). The concentration of HNE (arrow indicates the addition) was 16 μM. Data are presented as mean values ± SEM for 5-10 independent experiments. B. TMRE-stained cells were incubated for 2 h 30 min in DMEM medium with different glucose concentrations before addition of 16 μM HNE. TMRE fluorescence after 1h HNE treatment normalized to the fluorescence before HNE addition is shown. Data are presented as mean values ± SEM for 3 independent experiments. C. TMRE stained cells were starved for 2 h 30 min in 3.85 mM glutamine medium. TMRE fluorescence after addition of 2 μM oligomycin was normalized to the fluorescence without oligomycin. Data are presented as mean values ± SEM for 4 independent experiments.
