(a) The kinase activity of STK16 is not required for VSVG-GFP trafficking. HeLa cells were transfected with VSVG-GFP plasmids for 24 hours, then treated with DMSO, 1 μM or 10 μM STK16-IN-1 at 40 °C overnight. After shifted to 33 °C for 15 or 60 minutes, cells were fixed and stained by the GFP antibody to show the VSVG-GFP protein arrival at Golgi or PM, respectively. The right panel shows an enlarged view of boxed region in the left panel. (b) STK16 knockdown does not affect VSVG-GFP trafficking. HeLa cells were transfected with control or STK16 siRNAs for 48 hours before they were transfected with VSVG-GFP plasmids for 24 hours. Cells were incubated at 40 °C overnight, and shifted to 33 °C for 15 or 60 minutes, and then harvested for immunofluorescence using the GFP antibody. The right panel shows an enlarged view of boxed region in the left panel. Experiments were repeated two times and representative images are shown. Scale bar, 10 μm.
