NLM peptide induced peptide-specific cytotoxicity by T cells that recognizes NQM native sequence. T cells from a HLA-A*02:01⁺ donor were stimulated with either NQM (A) or NLM peptide (B ) for five times and the cytotoxicity was measured by standard ⁵¹Cr-release assay against T2 cells pulsed with NQM peptide. Similarly, CD3T cells from two different donors who are HLA-A*02:01⁺, were stimulated with analog NLM peptide for five rounds and the cytotoxicity was measured against WT1⁺ HLA-A*02:01⁺ leukemia cells 697, WT1- HLA-A*02:01⁺ leukemia cells SKLY-16, WT1⁺ HLA-A*02:01⁻ leukemia cell line HL-60 (C) and leukemia blasts derived from a HLA-A*02:01⁺ or negative patient with AML (D). Each data point represents the mean from triplicate micro-well cultures. Data represent results from seven experiments from three donors in panels A, B and C, and three experiments from three pairs of samples from AML patients.
