Figure 9. Expression of tissue specific proteins in thin sections of tumor tissue.

Figure 9

Four tumors were analyzed by immunohistochemistry for the presence of a panel of tissue specific proteins. All four samples were positive for a. the transgene encoded peptide PIWIL2 as well as the stromal markers for mesodermal derived tumors b. VIMENTIN and c. SAM. The neurogenic tumor neurocyte markers d. NESTIN and e. SYNAPOPHYSIN (SYN) were also detected in all samples. As were f. cytokeratin (CK) which marks tumors of epithelial origin, g. placental alkaline phosphatase (PLAP) that is a marker for reproductive cell tumors of germ cell origin and h. DESMIN, a myocyte marker for mycogenic tumors. In addition tumors expressed i. AFP, a fetal cell marker usually present in teratomas and j. epithelial membrane antigen (EMA) that marks tumors of epithelial origin. These data indicate that the tumors derived from PIWIL2 transfected human foreskin fibroblasts are poorly differentiated and do not preferentially express any markers associated with mature differentiated somatic cell types.
