The effect of siRNA-mediated targeted depletion of PIK3CA on apoptosis was evaluated by measuring the activity of caspases 3 and 7 in HCT116 A. and SW480 B. CRC cells. Briefly, cells were grown, transfected with PIK3CA siRNA and apoptosis determined by Caspase-Glo 3/7 assay (Promega) 72h after transfection. Controls included cells transfected with transfection reagent (mock ctrl) and Non-targeting siRNA. Data represent means ± SEM of at least triplicate experiments normalized to controls. PIK3CA siRNA was compared with Non-targeting siRNA. Ctrl, control. **, p< 0.01.
