A. OVCAR-3 cells transfected with empty (Sham), C124S mutant PTEN or G129E mutant PTEN HIS-tagged plasmids. The cells were incubated for 36 h post-transfection to allow the synthesis of transgenic PTEN and then cultured for further 24 h in a medium (EBSS) lacking growth factor (GF) and aminoacids (AA). Thereafter, the cell lysates were used for Western blotting to reveal the level of expression of transgenic PTEN (anti-HIS) and of active (pAKT at Thr308) AKT. A similar pattern of protein expression was observed in two other separate experiments. B.
