GFP Fluorescence following Ex Vivo Administration of AAV2-GFP ± WPRE in Human Retinal Tissue
(A) Transmission and fluorescence images were acquired on alternate days post-transduction with AAV2-GFP and AAV2-GFP-WPRE under standardized conditions. A non-transduced sample was imaged in parallel as a control. Normalized gray levels were calculated by subtracting the mean gray level for the control sample fluorescent image from that obtained from the equivalent image of transduced retina. (B) Normalized gray levels of two human retinal explants (patient 1, black circles; patient 2, gray circles) transduced with AAV2-GFP ± WPRE were plotted over time showing a trend for increased GFP expression in the presence of the WPRE.
