On day 5 and day 30 after co-culturing, 293T-B cells, CD40L-B cells and CD40L-sBAFF-B cells were harvested. a-c. B cells were co-cultured at an optimal ratio of 3:1 with CFSE-stained autologous CD8+ T cells. Cell divisions of CD8+ T cells were measured by CFSE dilution. d-g. B lymphocytes were cultured for 30 days, and then harvested, irradiated, washed, and co-cultured with purified autologous CFSE-labeled CD8+ T lymphocytes at an E: T ratio of 1:3 for one week. (d, e) Production of IFN-γ was measured by ELISPOT assay. (f, g) A representative profile indicating the percentage of CD8+ CD45RA CD62L+ CCR7+ central memory T lymphocytes after co-cultured with B lymphocytes. Data represent mean ± SD (error bars) for a representative experiment of three independent experiments. The paired t-test was used.
