The CFP-10_71–85 antigenic region is promiscuously restricted by M1 DR and DQ molecules. (A and B) Two CFP-10_71–85-specific CD4 T-cell lines were generated from animal 128-15 (A) and animal 129-15 (B). ICS assays were conducted to determine the peptide specificity for each T-cell line. The percentage of CD3⁺ CD4⁺ cells positive for IFN-γ and/or TNF-α for each sample was determined. (C and D) ICS assays were performed by using RM3 cells or RM3 cells stably transfected with the indicated MHC class II alleles associated with the M1 MHC haplotype to determine the restriction for animal 128-15 CD4 CFP-10_71–85 (C) and animal 129-15 CD4 CFP-10_71–85 (D). Cells were gated on CD3⁺ CD4⁺ CD8⁻ events, and the percentage of IFN-γ-positive TNF-α-positive cells was determined.
