“Non-target OTUs” correspond to OTUs that did not match the COI sequence of a species included in the mock sample. Each point represents data for one sequencing run for one non-target OTU, so that for example, four non-target OTUs found in all seven indexed PCR trials in sequencing run 1 and two non-target OTUs found in all seven PCR trials in sequencing run 2 had a total number of reads >10 for both sequencing runs.
