Far-UV CD spectra (185–260 nm) of 1 μM T-peptides, TolC trimer, and their complex at a 1:1 molar ratio were measured in cuvettes with optical path lengths of 0.1, 0.2, and 1.0 mm. Spectra of peptides and difference spectra of TolC-bound peptides (“TolC/T-peptide” – “TolC”) expressed in mean molar residue ellipticity units were analyzed using Selcon3, Contin, and K2D programs on the DichroWeb Internet site.42 The buffer consisted of 0.1 M sodium phosphate (pH 7.0) and 0.03% DDM.
