In vitro transcription coupled with translation was performed using a TNT-coupled reticulocyte lysate kit (Promega) as described in Materials and Methods. The translation products were separated by a 4–15% gradient SDS-polyacrylamide gel, and the gel was treated with Amplify (Amersham Life Science), dried, and exposed to Kodak BioMax MR film. The predicted molecular weights of mSIG-1, mSIG-1A, mSIG-1B, mSIG-1C, mSIG-1D and mSIG-1E are 25.3, 21.6, 24.5, 21.2, 13.0 and 17.9 kDa, respectively. The representative gel from two experiments was shown.
