RNase III cleavage of in vitro transcripts. Five micrograms of in vitro transcribed RNA was incubated with purified RNase III and activated with MgCl2 at 37°C for 15 min. Five hundred nanograms of RNA was removed before the addition of enzyme and MgCl2. Samples were run on a denaturing polyacrylamide gel and imaged with ethidium bromide.
