(A) SH87 cell routinely maintained in glucose-only PFCDM was sub-cultivated into PFCDM with 4 g/l glucose, or 4 g/l glucose +3 g/l maltose. On Days 2, 4, 6 and 8, cell-free conditioned medium (CM) were harvested from both cultures. Viable cell densities (lined marker) and culture viabilities (marker only) were also monitored. (B) CM from the maltose supplemented culture were incubated for a further 3 days at 37 °C. Samples were harvested daily from these CM, starting from the day of harvest, to determine glucose and maltose concentrations. (C) CM harvested from the glucose culture were spiked with maltose to a final concentration of 3 g/l prior to further incubation at 37 °C. Samples from these CM were harvested daily for a further 3 days to determine glucose and maltose concentrations. The averages and standard deviations from 2 replicate shake flasks were plotted.
