(a) Interaction assay of CTB4a^KD and AtpB in yeast (strain AH109). Interaction was determined by a growth assay on medium lacking Trp-Leu-His-Ade. (b) GST-pull down assay confirming the interaction between CTB4a^KD and AtpB in vitro. Purified CTB4a^KD-GST and AtpB-His fusion protein from E.coli BL21 were pull-downed by GST beads and blots were probed with anti-GST or anti-His. (c) Interaction assay of CTB4a^KD and AtpB in vivo. Co-expressed CTB4a^KD-HA or HA and AtpB-Myc in tobacco leaves were immunoprecipitated by anti-HA antibody and blots were probed with anti-Myc or anti-HA.
