Effect of temperature, pH level, and ionic strength on the binding capacity of GFP-CBM fusions to Sigmacell 20 cellulose powder. A, adsorption percentage was determined in 100 mm phosphate buffer, pH 7, after contacting 50 pmol of GFP-CBM with 5 mg of Sigmacell 20 for 20 min at 4 °C and 90 °C. B, adsorption percentage was determined in 100 mm phosphate buffer, 25 °C after contacting 100 pmol of GFP-CBM with 5 mg of Sigmacell 20 for 20 min at pH 5 and pH 9.5. C, adsorption percentage was determined at 25 °C after contacting 50 pmol of GFP-CBM with 5 mg of Sigmacell 20 for 20 min in 100 mm and 1 m phosphate buffer, pH 7. Experiments were performed in triplicate.
