HEK293 cells were transfected with either vector alone (pcDNA3) or pcDNA3-ACE2-Fc. Cell lysate and culture medium were separately collected and were subsequently subjected to western blot (A and B) and the phenylalanine assay using Ang II substrate (C and D). Full-length blots/gels are presented in Supplementary Figure S3. ACE2 activities were detected and these activities were completely inhibited by MLN-4760. The results in C and D are expressed as the means ± SD of three independent experiments.
