Stress responses mediate HAS2 deficiency-induced senescence. Fibroblasts were transfected with HAS2 siRNA or control siRNA. Cell lysates were harvested at various time points after transfection and subjected to Western blots. The expression of proteins involving in DNA damage response pathway including phospho-H2A.X (A), phospho-p53 (B), and proteins involving in stress response pathway including phospho-perk (C), phospho-elF2α (D), and Bip (E) were analyzed. Densitometry quantification for phospho-perk was included in C. The experiments were performed three times.
