Kinetics of synthesis of DNA by WT virus following infection of P4 cells. Viral stocks were prepared by infecting MT4 cells with virions produced by transfected HeLa cells and collecting culture medium on the day of peak virus production. The virions were treated with DNase I and used to infect P4 cells by spinoculation. The cells were then washed and incubated at 37°C. After various times of culture, DNA was extracted from the cells, and DNA corresponding to sequences in the U5, Env, and Gag regions was quantified by real-time PCR. (A) Kinetics of synthesis of viral DNA in unpretreated P4 cells. (B) Kinetics of synthesis of viral DNA in P4 cells that had been pretreated with 250 μM hydroxyurea for 18 h prior to infection.
