Excess aspartate exacerbates SMM-induced starvation. BY4743 wild-type cells and gcn4Δ/gcn4Δ cells treated as in Figure 3A, except that they were plated onto SC, SC-ile-val with 2 µg/ml SMM (top panels), SC with 2 g/liter aspartate added, and SC-ile-val with 2 µg/ml SMM and 2 g/liter aspartate added (bottom panels). The BY4743 cells are unaffected by addition of excess aspartate in the SC medium. When starved for isoleucine and valine, the growth of the BY4743 strain is inhibited, and the growth is made worse by the presence of the excess aspartate. The gcn4Δ/gcn4Δ strain is included as a control to show that the SMM is working.
