Domain architecture of RIG-I: Schematic representation of the RIG-I domains. RIG-I is comprised of four major domain groups including the CARDs (black), and RNA-stimulated ATPase core (green), a helical regulatory and binding domain inserted into the second lobe of the ATPase called HEL2i (cyan), and a triphosphate recognition and RNA binding domain at the c-terminus annotated the CTD (orange). Hel1 of the ATPase core contains four conserved motifs, motifs I, II, III and the Q-motif. Residues in these motifs are necessary for proper RIG-I function, and the sites of disease related mutants, E373 and S411, are indicated (*).
