Model schematic. The metabolic network is comprised of 46 metabolites interacting through 53 enzymatic reactions. The major pathways involve glycolysis, glutaminolysis, the TCA cycle, the PPP, and shuttle reactions between mitochondrial (shaded rectangle) and cytoplasmic compartments. The abbreviations for the metabolites, cofactors, and enzymes are given in Supplementary File S3. The colored nodes represent the metabolites for which the fold-change has been measured experimentally during the knockdown of enzyme GOT1 (shown in red). The arrows represent the direction of the reaction fluxes in the baseline model at the initiation of the simulation.
