FIGURE 4:

Microinjected HPCD and STARD4 overexpression enhances bidirectional sterol transport. HPCD and rhodamine–dextran were microinjected immediately before labeling of cells with DHE. U2OS-SRA cells were transfected with GFP-STARD4 for 18 h before the experiment. (A) FRAP measurements for single cells microinjected with HPCD or transfected with GFP-STARD4. Control, t1/2 = 14.5 ± 1.5 min; HPCD (1 × 109 molecules/cell), t1/2 = 1.3 ± 0.3 min; GFP-STARD4, t1/2 = 4.3 ± 0.5 min. (B) DHE efflux curves for cells microinjected with HPCD or transfected with GFP-STARD4. Control, t1/2 = 14.9 ± 0.9 min; HPCD (0.5 × 109 molecules/cell), t1/2 = 4.1 ± 0.4 min; GFP-STARD4, t1/2 = 4.9 ± 0.6 min. Each data point is derived from an average of at least 10 experiments (±SE). Data are fitted to single-exponential curves.
