Pds1p remains high in the spc110-226 arrest. Control SPC110 (JBY24) and spc110-226 (JBY60) diploid strains were arrested at 22°C with 0.1 M HU. After 1.5 generations, cells were released to fresh YPD at 37°C where spc110-226/spc110-226 cells arrested as large budded cells and control cells cycled. Total cellular protein extracts were collected at times indicated after release to 37°C. A Western blot against Pds1p-HA is shown. Pds1p runs as a doublet. The same amount of total protein was loaded in each lane as determined from a Coomassie Blue-stained polyacrylamide gel of the same samples. The negative control (N) is from the untagged strain JBY11.
