miR-718 expression is induced by LPS and N. gonorrhoeae and is dependent on IRAK1. A, immortalized mouse BMDMs from WT and Irak1/miR-718-dKO were stimulated with LPS (100 ng/ml) for 2 h or cultured in medium alone. B, immortalized BMDMs from WT and Irak1/miR-718-dKO were infected with various m.o.i. of N. gonorrhoeae or LPS. C, immortalized microglia were stimulated with LPS (100 ng/ml) for 2 h or cultured in medium alone. D, mice were injected with 1 mg/kg body weight LPS or PBS (5 mice/group). After 4 h, spleens were harvested. RNA from the cells in A–D was extracted, and miR-718 levels were quantitated by TaqMan qPCR. miR-718 levels are shown relative to levels of small non-coding RNA (sno202). The data shown represent the mean ± S.D. of triplicate determinations and are representative of three independent experiments with similar results.
