miR-718 represses the production of proinflammatory cytokines in macrophages.
A–F, stable macrophage cell lines expressing miR-718 or the corresponding empty vector control (Con miR) (A, C, and E) and Anti-miR-718 or the corresponding empty vector control (Con Inh) (B, D, and F) were stimulated with 100 ng/ml LPS at various time points. RNA was extracted, and miR-718 levels were quantitated by TaqMan qPCR (A and B). IRAK1 levels were quantitated by SYBR Green qPCR (C and D). ELISA was performed to measure TNFα release (E and F). The data shown represent the mean ± S.D. of triplicate determinations and are representative of three independent experiments with similar results. G, Western blotting was performed with lysates from Con miR and miR-718 macrophages stimulated with 100 ng/ml LPS for 2 h or medium (Med). ns, not significant.
