Figure 7. — Interaction of GRK-2 with AMX-2. A, quantitative RT-PCR analysis indicates that the mRNA level of amx-2 is not significantly changed in the grk-2 mutants compared with N2. Results are from four experiments each done in duplicate. B, co-immunoprecipitation experiments demonstrate that GRK-2 interacts with AMX-2, and this interaction is unaffected by G protein activation with aluminum fluoride. No co-immunoprecipitation is observed in the amx-2(ok1235) and grk-2(gk268) mutant strains. C, top panel, AMX-2 was immunoprecipitated from N2, amx-2(ok1235), grk-2(gk268), and grk-2(rt97) lysates and then analyzed by Western blotting using a mouse monoclonal phospho-Thr antibody. The lysates were also analyzed for GRK-2 (middle panel) and AMX-2 (bottom panel) expression. This result is representative of five experiments. All error bars indicate S.E.
