Dose-response of BRF110. The EC₅₀ of BRF110 was measured in a cellular assay in SHSY5Y cells cotransfected with a DR5-driven luciferase reporter plasmid and CMV-driven Nurr1 and RXRα plasmids. A CMV β-galactosidase plasmid served as a transfection control. Varyious concentrations of BRF110 were added to the culture medium at 24 h after transfection. Cells were harvested 24 h later, and activation of the Nurr1:RXRα heterodimer was determined by luciferase assays (Promega).
