ERD10 is not detectable by NMR when overexpressed inside plant BY-2 cells. The 1H-15N-HSQC spectrum of 100 μM ERD10measured in 50 mM MES buffer pH = 6.5 that is represented by the red cross-peaks is overlayed with 15N-labeled glutamine and asparagine side chain signals that are represented as blue contours (A). The 1H-15N-HSQC spectrum of BY-2 cells transformed with erd10 under the control of an inducible promoter does not display visible protein cross-peaks after estradiol-induced protein expression and continued growth for 48 h (B). Only the 15N-labeled glutamine NH2 side chain signals are detected, which indicate that the 15N-source is successfully metabolized into amino acids.
