Aptamer induced cell growth inhibition. (A) Predicted G-quadruplex structures with two flat planes in aptamer HL-1. (B) Biostability assay. Aptamer HL-1 was incubated in culture medium containing 10% human serum at 37°C, and residual products were measured by gel electrophoresis at different time points. (C) Cell proliferation analysis. Cells were treated with aptamer HL-1 at different final concentrations or synthetic ssDNA library as control, and changes in cell growth were monitored by MTT proliferation assay. The aptamer induced significant growth inhibition of Maver-1 cells 3 days post treatment, but had no effect on off-target Jeko-1 cells.
