A. 3D-sphere formation. MCF7 cells were plated in low-attachment cell culture plates (6-well format), to assess the ability of CAPE to inhibit mammosphere formation. Note that CAPE treatment was sufficient to significantly reduce mammosphere formation, with an IC-50 of approximately 2.5 μM. B. Cell viability. The effects of CAPE on the viability of adherent cells (MCF7 cells and hTERT-BJ1 fibroblasts) was assessed in parallel, using the SRB assay, in a 96-well format. Note that CAPE significantly reduces the viability of MCF7 cells, with an IC-50 of 10 μM. Similar results were obtained with hTERT-BJ1 cells. Cell viability was assessed after 72 hours of treatment.
