Lepr and Insr expression in hypothalamic POMC neurons. (A) We detected 30 Lepr and 83 Insr expressing cells out of the 163 cells in our sequencing cohort. (B) The percentages were compared to the number of cells that exhibited pStat3 and pAkt immunoreactivity post-administration of leptin and insulin (respectively). (C) Representative images showing POMC-eGFP, immunoreactivity of pStat3 (upper panel) and pAkt (lower panel). Cells that were double-stained were marked with white arrows. (D) The t-SNE plot shows a lack of clear clustering for Lepr and Insr expressing cells, indicating that expression of either receptor alone was not sufficient to drive segregation at the whole transcriptome level.
