Distribution of theDrosophilaCTPsyn wild-type and mutant proteins in cells co-expressing the constructs. Wild-type CTPsyn-mCherry (A′-E′) was transfected along with Venus-tagged CTPsyn mutant proteins CTPsynΔ20 (A), CTPsynΚ2Ε (B), CTPsynY3E (C), pyrG/1−22CTPsyn (D) and pyrG/N4KCTPsyn (E), and their distribution in the cytoplasm monitored. While CTPsynΔ20, CTPsynΚ2Ε , CTPsynY3E and pyrG/1−22CTPsyn proteins assembled into aggregates that did not overlap with CTPsyn cytoophidia (A′′-D′′), pyrG/N4KCTPsyn co-assembled with the cytoophidia (E′′). DNA staining with Hoescht is shown in blue and the Venus- and mCherry-tagged proteins are in green and red, respectively. Scale bar=5 µm.
