Interaction of yeast ribosomes with Stx1 (magenta), Stx2 (blue) and ricin (green) holotoxins. Toxins were immobilized on a CM5 chip. Ricin was immobilized on Fc2 at 1873 RU, Stx1 was immobilized on Fc3 at 2541 RU, and Stx2 was immobilized on Fc4 at 1853 RU. Fc1 was activated and blocked as a control. Ribosomes were passed over the surfaces at 2.5, 5, 10, 20 and 40 nM. The running buffer contained 10 mM Hepes, pH 7.4, 150 mM NaCl, 5 mM MgCl2, 50 µM EDTA and 0.003% Surfactant P20. The surface was regenerated with one-minute-injection of 3 M NaCl for three times. The interaction was measured at 25 °C. The signals were normalized to the ligand level of 1000 RU. The experiment was replicated three times.
