Impact of exchanging the entire envelope gene or just the S region between clones of genotype B and genotype C on hepatitis B virus (HBV) DNA replication and virion secretion. Both the 1.1mer construct (left panels) and SphI dimer (right panels) were used for the exchange of the entire envelope (env) gene or just the S region. The original and chimeric constructs (2 µg) were transfected to Huh7 cells, which were harvested four days later. (A) Southern blot analysis of intracellular replicative DNA (B) and extracellular virion-associated DNA from a representative transfection experiment. Mixed probes of genotype B and genotype C were used for hybridization. Positions of the 3.2-kb, 2.2-kb, and 1.0-kb DNA are indicated.
