Amidation of RPRFa is essential for high-affinity potentiation of ASIC3 currents. Representative current trace illustrating the lack of potentiation of ASIC3 current by 50 μM RPRF, lacking the C-terminal amidation. In contrast, RPRFa (50 μM) potentiated ASIC3 currents. Peptides were applied in the conditioning period at pH 7.4 for 1 min (red bars). ASIC3 was activated by pH 6.3 (black bars).
