Control of IRGM knockdown in Huh7.25CD81 cells. Huh7.25CD81 cells were treated with two different siRNAs against IRGM or Control siRNA. (A) Total protein levels were examined by immunoblotting with an IRGM antibody. The migration of the molecular weight markers is presented to point out that the anti-IRGM antibodies specifically recognize IRGM at its expected position. The specificity of the siIRGMs was verified, because they silenced only the target gene and did not affect the intensity of nonspecific bands. (B) Intracellular staining of endogenous IRGM. Cells were fixed with 4% PFA, blocked, and immunostained with an antibody against IRGM.
