Occludin protein expression was determined on cell lysates collected from RBMECs treated with α-1-acid glycoprotein (AGP) for 3, 6, 12, 24, 48 and 72 hrs, using Western blot analysis. AGP treatment at 50 μg/ml (horizontal bars) and 500 μg/ml (diagonal line) were compared to control monolayers (open bars). Occludin protein expression was normalized by β-actin expression with representative blots shown above the bar graph. Statistical analysis was done using one-way ANOVA with Student Neuman–Keuls post-hoc analysis. * indicates p-value <0.05; ** indicates p-value <0.01; *** indicates p-value <0.001 when compared with respective control; n = 4–9. Two-way ANOVA results indicate that occludin protein levels are significantly different following 50 μg/ml AGP treatment versus 500μg/ml treatment at 6, 12 and 72 hrs.
