Poly(I:C)-induced testicular innate antiviral response in vivo. A and B, Expression of inflammatory cytokines and antiviral proteins in vivo. A mixture of poly(I:C) and lipofectamine RNAiMAX was locally injected into one testis of 5-week-old TLR3−/− mice, whereas lipofectamine RNAiMAX alone was injected into the other testis (control) of the same mice. At 6 hours after injection, total RNA was extracted from the testes, and relative mRNA levels of the cytokines (A) and the antiviral proteins (B) were analyzed by real-time qRT-PCR. C, Induction of antiviral proteins at protein levels in vivo. The testes were injected as described above. At 24 hours after injection, the testicular lysates were subject to Western blot to probe the antiviral proteins using specific antibodies. Images are representatives of at least 3 experiments.
