Mapping Su(Hw) ZF recognition of binding modules within the SBS motif. (A) Top: quantification of binding properties of Su(Hw)WT and Su(Hw) ZF mutants to DNA probes carrying mutations in either the upstream core (mU), the central core (mC) or the downstream core (mD) mutant probes. For each of these probes, five nucleotides in the core were mutated away from the consensus sequence. Indicated is the percent bound probe observed using 0.3 μg (light gray) or 1.0 μg (dark gray) of Su(Hw) protein. For comparison, data are shown using the PM probe (light blue, Figure 3). Data shows the average of two experiments. Bottom: shown is a schematic of Su(Hw) binding with a full consensus SBS, showing which ZFs are involved in recognition of specific cores.
