Evaluation of LY2157299 for acute cytotoxicity against individually cultured TDE_CC and TDF_SM cells. Cytotoxicity was assessed using the CytoTox96 Non-Radioactive Cytotoxicity Assay (Promega Corp., Madison, WI) as a function of lactic acid dehydrogenase (LDH) released into the culture medium measured at 1 and 3 hours after their initial exposure to LY2157299 in dimethyl sulfoxide (DMSO) at the indicated concentrations. Each well was assayed in triplicate with an initial cell plating density of 15 × 10³ cells/well in 96-well culture plates coated with collagen type I. Blank wells did not contain cells, and vehicle control cultures were treated with 10 μL DMSO in culture medium without LY2157299 (0 concentration). Indicated values represent the relative to maximum LDH release produced by treatment with lysis buffer provided with the assay kit. Note under these conditions that the LY2157299 treatment is not acutely toxic to either the cultures of TDE_CC (A) or TDF_SM (B) cells. Data are expressed as means ± SD.
