Heat‐shock protein 90α (hsp90α) was not released in response to inducible nitric oxide synthase (iNOS) activity. Human βLox5 (a) and human islets (b) were treated for 24 hr with media alone (Ctrl) or with interleukin‐1β (IL‐1β), tumour necrosis factor‐α (TNF‐α) and interferon‐γ (IFN‐γ) (Cyt). NOS2 expression was detected only after cytokine treatment as measured by quantitative RT‐PCR (UD, undetected). Human βLox5 cells were treated for 6 hr at 37° with medium alone or 100 μm 1400W, an iNOS inhibitor, followed by 24 hr treatment with pro‐inflammatory cytokines. Following these treatments, extracellular hsp90α levels were detected by ELISA. hsp90α release by beta cells was increased in response to cytokine exposure irrespective of iNOS inhibitor addition (c). Data are presented as relative to control cells cultured without cytokines or 1400W. Extracellular hsp90α values detected ranged between 10·0 and 34·6 ng/ml.
