(a) Thermostability of PolB1-HE and sub-assemblies determined by DSF analysis with 40 μM protein in the presence of SYPRO Orange. Fluorescence at emission wavelength 550 nm was recorded in increments of 1 °C. Data are plotted using Kaleidagraph version 4.5. (b) PCR activity of PolB1 and the indicated complexes. Reactions contained 100 nM of each enzyme. The ability to generate a PCR product (∼450 bp) was analysed by electrophoresis on a 1% agarose gel. A DNA ladder is on the left with indicated sizes.
