STAT3 is required for IL-6-mediated repression of G6Pase, and STAT3 represses PEPCK in the absence of IL-6. HepG2 cells were transfected with control or STAT3-specific siRNA. Whole-cell extracts were prepared and analyzed for STAT3 expression (A). siRNA-transfected cells were placed in OptiMEM for 16 h and then treated with IL-6 (20 ng/ml) or forskolin (10 μm) or both.
