Kinetics of CREB Phosphorylation within Cumulus Cells of COCs in Response to FSH or N6-cAMP Plus 8-Br-cAMP and the Effect of KG-501 on Oocyte Maturation

Cumulus cells from 50 COCs were collected at various time point for immunoblotting with anti-phos CREB and anti-CREB antibodies.COCs were cultured in HX medium with or without 50 IU/liter FSH (A). COCs were cultured in HX medium containing 50 IU/liter FSH (B) or pulsing with 250 μm N6-cAMP and 8-Br-cAMP for 3 h (C). Different concentrations of KG-501 (0–25 μm) were added when mouse COCs were cultured in HX medium containing 50 IU/liter FSH or pulsing with 250 μm N6-cAMP (N6) plus 8-Br-cAMP (8-Br). The percentage of GVBD was recorded at 24 h of culture. Columns with different letters were significantly different (P < 0.05) (D).
